چکیده
Background: CD4+ T cells with cytotoxic functions (CD4+ CTL) have gained attention in recent years. Accumulating evidence supports their importance defense against human viral infections such as CMV 1 , EBV 2 dengue 3 HIV 4, 5 and SARS-CoV-2 6 . Moreover, expansion of so called CD28null been reported the blood patients rheumatic diseases rheumatoid arthritis (RA) 7 myositis 8 vasculitis 9 well cardiovascular 10 Objectives: Here, we aimed to investigate presence clonal CTL peripheral (PB) synovial fluid (SF) RA using single cell technologies. Methods: We assessed expression effector molecules transcription factors (n=21) paired (n=16) from ACPA- APCA+ by multi-parameter flow cytometry. performed sequencing, combination 5´ TCRab on purified ACPA+ (n=7). Results: Flow cytometry experiments show that Granzyme-B+ Perforin-1+ are significantly increased SF compared (p=0.0072). The could be confirmed sequencing each patient tested found adhesion G-protein coupled receptor GPR56 is selectively expressed recently described helper (T PH ) T-cell subset 11 associates tissue resident memory markers LAG-3, CXCR6 CD69. In blood, a previous report 12 showing delineates cells. Finally, expanded TCR clones expressing were identified and, for some patients, corresponding blood. Conclusion: marker residency receptors. highlights suggests potential therapeutic target (Figure 1). References: [1]Casazza J. P. et al ., J Exp Med 2006, 203 (13), 2865-77. Somatic mutations in clonally expanded cytotoxic T lymphocytes in patients with newly diagnosed rheumatoid arthritis
Somatic mutations contribute to tumorigenesis. Although these mutations occur in all proliferating cells, their accumulation under non-malignant conditions, such as in autoimmune disorders, has not been investigated. Here, we show that patients with newly diagnosed rheumatoid arthritis have expanded CD8+ T-cell clones; in 20% (5/25) of patients CD8+ T cells, but not CD4+ T cells, harbour somati...
متن کاملResistance to apoptosis and elevated expression of Bcl-2 in clonally expanded CD4+CD28- T cells from rheumatoid arthritis patients.
Patients with rheumatoid arthritis have a subset of CD4+ T lymphocytes that are characterized by a defect in CD28 expression. CD4+CD28- T cells frequently undergo clonal expansion in vivo. These clonotypes include autoreactive cells and persist over many years. The clonogenic potential and longevity of these T cells could be related to an altered response to apoptosis-inducing signals. To explo...
متن کاملKiller cell activating receptors function as costimulatory molecules on CD4+CD28null T cells clonally expanded in rheumatoid arthritis.
Expansion of CD4+CD28null T cells is a characteristic finding in patients with rheumatoid arthritis. Despite lacking CD28 molecules, these unusual CD4 T cells undergo clonal proliferation and form large and long-lived clonal populations. They produce high levels of IFN-gamma, exhibit autoreactivity, and have cytolytic function. The mechanisms facilitating the expansion and longevity of CD4+CD28...
متن کاملCytotoxic CD4 T Cells in Antiviral Immunity
CD4 T cells that acquire cytotoxic phenotype and function have been repeatedly identified in humans, mice, and other species in response to many diverse pathogens. Since CD4 cytotoxic T cells are able to recognize antigenic determinants unique from those recognized by the parallel CD8 cytotoxic T cells, they can potentially contribute additional immune surveillance and direct effector function ...
